Increased vascular permeability is one of the characteristics of diabetic patients. The endothehal cells regulate the vascular permeability. We studied the vascular permeability using albumin. We intended to make a model of albumin transcytosis, and to reveal the mechanism of glucose-induced albumin transcytosis. Endothelial cells were cultured on collagen-coated microporous filter to make an endothelial cell monolayer on the filter. This monolayer was placed between the upper well and the lower one. To estimate albumin transcytosis from the upper wells (luminal sides) to the lower wells (basolateral sides), we added Evans' blue dye labeled albumin (EBA) in the upper wells and measured the amount of EBA in the lower wells. The amounts of EBA in the lower wells increased with the hours of incubation at 37 ℃, but not at 4 ℃ Two or 3 days' incubation with high glucose concentration (20.5 mM) increased the amounts of EBA in the lower wells significantly. Incubation with phorbol myristate acetate (PMA) also increased the amounts of EBA in the lower wells. This may suggest that high glucose levels increased albumin transcytosis through the activation of protein kinase C. This glucose-induced albumin transcytosis may explain in part, the increased vascular permeability in diabetic patients.
