Optical methods for monitoring membrane potential with fast voltage-sensitive dyes have been introduced as a powerful tool for studying cardiac electrical functions. These methods offer two principal advantages over more conventional electrophysiological techniques. One is that it is possible to make optical recordings from very small cells which are inaccessible to microelectrode impalement, and the other is that multiple sites/regions of a preparation can be monitored simultaneously and provide spatially-resolved mapping of electrical activity. The former has made it possible to record spontaneous electrical activities in early embryonic pre-contractile hearts, and the later has been applied for the mapping of the propagation patterns of electrical activities in the cardiac tissue. In the course of optical studies of the excitation spread pattern of the vertebrate heart, we employed multiple-site optical recording of membrane potential activity, using a voltage-sensitive merocyanine-rhodanine dye (NK 2761) and a multiple-element photodiode array, to monitor action potentials in an isolated rat atrial preparation. We optically mapped the normal excitation spread pattern from the physiological pacemaker. We also mapped the excitation spread pattern during the events of tachycardia-like excitation (TE) evoked by electrical stimulation. These maps indicate that, during the event of TE, the excitatory waves were often rotated around the blocked area including ostium of the superior vena cava. The optical mapping during the initiation phase of TE indicated that the decrease of the conduction velocity and the appearance of blocked areas contributed the establishment of TE.
