The enzyme activity was maximum at pH 8.0 and at 60℃. The purification of the enzyme was carried out by being adsorbed onto insoluble collagen (30℃, 60min), centrifuged, and eluted with 1M sodium cloride solution. Rate of adsorption of the enzyme was maximum, 80% for total activity, when ratio of insoluble collagen to the enzyme was 300 : 1,and the elution also 80% for activity of the enzyme adsorbed. Further purification of the enzyme fraction by Sephadex G-75 and G-150 column chromatography was found to be a 6.5% over all yield and a 1.13 fold increase in specific activity.
