17580247","subitem_description_type":"Other"},{"subitem_description":"平成17年度～平成19年度科学研究費補助金(基盤研究(C))研究成果報告書","subitem_description_type":"Other"},{"subitem_description":"研究概要:(平成18年度時点)これまでの我々の研究で,精子ヒアルロニダーゼに対する強い抗ヒアルロニダーゼ効果は,透明帯へ結合した精子で引き起こされる先体反応を抑制する作用機序を介してブタ体外受精時の多精子侵入の頻度を効果的に減少させることを明らかにした。一方,ブタ卵子で多発する多精子侵入は,表層顆粒の開裂に伴った一連の透明帯(ZP)反応の不備に起因していると思われる。そこで本研究は,体外成熟ブタ卵に直流電気パルス刺激(ES)を与え,卵活性化に伴うZP構成糖タンパク質(ZP1,ZP2,ZP3ならびにZP4)の糖鎖末端の変化を追究した。まず初めに,ES処理卵におけるプロテアーゼ溶解反応へのZP抵抗性ならびにZPへの結合精子数の変化を経時的に観察したところ,ES処理4時間後に透明帯の硬化が起こり,次いでES処理12時間から18時間に掛けて結合精子数の著しい減少が認められた。そこで,ブタにおける精子-ZP間の結合に関与する ZP構成糖タンパク質の塘鎖末端に対して特異的な親和性を示すWGA, S-WGAおよびRCA-1の各レクチンを用い,ZP構成糖タンパク質へのレクチン結合性変化をSDS-PAGE後のECL法で検出した。その結果,GlcNAc末端を認識するWGAとS-WGAはZP2ならびにZP3へ結合し,これら結合能はES処理18時間以降に有意に低下した。一方,GalNAc末端を認識するRCA-1では,ZP1,ZP2およびZP3への結合が検出され,ES処理4時間後にZP1への結合能が,12時間後には
