The author studied cornified reaction of epithelial cells of vagina of rats, by injecting (1) estrone into subcutaneous tissue of body and (2) small amount of it into vagina. To obtain 50% of possitive reaction, 4.μg of Estrone was necessary in the case of subcutaneous injection and 0.03ug in the case of intravaginal injection. Intravaginal injection was about 130 times more sensible than subcutaneous injection, and it was found that the former is of value as a method of measuring estrogen in the blood stream. 2. The author measured the amount of Estrogen in the blood stream of rat in the morning during the period of proestrus by making extraction samples of blood. The extraction samples was made by removing fat in addition to Szego and Robert method and small amount of it was injected into vagina.
