In this study, we examined the expressions of G1 cyclins and cyclin-dependent kinases (cdks) in the rat hippocampus after transient ischemia. Transient forebrain ischemia was induced in rats by 4 vessel occlusion for 10 min, and then restored through the carotid arteries for 3, 6, 12, 24, 48 and 72 h. The expressions of cyclin D1, cyclin E, cdk4, and cdk2 genes were analyzed by RT-PCR. The expressions of cyclin D1, cdk4 and cyclin E mRNA gradually increased from 6 to 48 h after ischemia. Immunohistochemically, D-type cyclin was detected in the pyramidal cells of the hippocampus after transient ischemia. After 48 h, the cells in CA1 stained with anti-cyclin D1 antiserum were more intense than those in CA3. The nuclei of these cells were deeply stained. These results demonstrate that cell cycle regulatory mechanisms persist even in differentiated neurons and could be induced by ischemic insult.
