By examining the increasing curve on the acid soluble material and hyperchromicity, the hydrolytic process of DNA was limited by DNase I under very low Mg^<2+>concentration. The partially hydrolyzed product (ph-DNA) did not show any spectral changes by the disruption of hydrogen bonds involving the reactive amino acid. Therefore, ph-DNA is considered to still retain a double helical structure. ph-DNA was obtained from the reaction mixture under hydrolytic condition and when more Mg^<2+> was added to the reaction mixture, the b-form DNA was produced.
