Micro fluorospectrophotometry, using Nikon SUR -F type, equipped with high pressure mercury lamp, (HBO Osram 200W Toshiba)was done on Mf. immitis, earthworm epidermis, mouse retina, carrot, authentic Vitamin A palmitate and authentic flavins. Mf. immitis showed microscopic fluorescence-spectra with Fmax at 540 nm, excited both by 365 and 410nm. Authentic flavins, riboflavin, FMN and FAD, gave fluorescence spectra with F max between 530 and 540 nm, excited by 365nm, while they did not fluoresce when excited by 410nm. Similar Autofluorescent granules were seen in the earthworm epidermis (head segments) besides strong red fluorescence of porphyrins in the deeper parts of the body. Microscopic fluorescence-spectra were obtained in the fluorescent parts in the epidermis (head segment), with F max neat 540nm, excited by 365nm, and with F max near 560nm with shoulder between 580 and 590 nm. excited by 410 nm. Similar auto fluorescent granules were observed in the outer rod segment of mouse retina and fluorescent parts showed fluorescense spectra with F max 525 nm excited by 365 nm Vitamin A palmitate showed fluorescence-spectra with F max at 550 nm, excited both by 365 and 410 nm. Sections of carrot, containing betacarotene, gave similar spectra with F max at 540 nm, excited both by365 and 410nm. Very similar fluorescence-spectra were obtained, using, newly introduced microfluorospectrophotometer, in our University Hospital, with two F max at 510 and 550 nm excited about 400 nm, in the earthworm epidermis and in the outer rod segment of retina of Asian toad.","subitem_description_type":"Other"},{"subitem_description":"2. Ultramicrospectrophotometry, using UMSP-1 Zeiss, was done in the earthworm epidermis and frog retina. Ultramicroscopic absorption spectra were obtained in the fluorescent granules in the earthworm epidermis, with $ \\lambda $ max between 410 and 420 nm. In the fluorescent granules in the outer rod segment of frog retina, $ \\lambda $ max was between 420 and 430 nm. Absorption spectrum could not be obtained in case of microfllanae. Fluorescence recorder (Farrand Optical Co. UVIS) was atouched to UMSP-1 Zeiss and excitation was done by monochronomatic light, using grating monochromator. Ultramicroscopic-fluorescence spectra were compared among the autofluorescent granules in Mf. immitis. earthworm epidermis and in the outer rod segment of frog retina. The spectra showed F max at 525nm with a shoulder at 550nm in case of Mf. immitis, excited by 365 nm. Those of the earthworm epidermis gave F max at 530 nm with a shoulder at 570 nm, excited by 365 nm and at 535 nm with a shoulder at 580 nm, excited by 410nm. F max were seen at 530nm with shoulder at 580nm excited by 365nm, and at 540nm with shoulder at 580 excited by 410nm, in the fluorescent granules in the segment of frog retina. Vitamin A paltimate gave F max at 530 and carrot gave F max at 520nm. 3. Histochemical demonstration of Vitamin A was done using Wake's modified method of Kupffer's gilding with chloroauric acid (HAucl_4・4H_20) Black positive granules were detected in all microfilaliae, examined without previous irradiation. The inhibitory effect of UV-irradiation againist this reaction made it difficult to prove the identity of such black granules (Vitamin A) with the autofluorescent granules in the mifilariae. However, in the Mf. irradiated comparatively shorter time, were detected red granules (weak Vitamin A reaction) in the same locality with autofluorescent granules. Black granules were also detected in the fluorescent parts of the earthworm epidermis in the head segment and in the outer rod segment of frog retina.
