As a result, phylogenetic tree from hemoglobin data resembled tree from the mitochondria DNA data and DNA hybridization. These two trees were reversed the relationship between California quail and Guinea fowls. In parallel of these study, we developed the PCR application for the rare species which difficult to get the blood. We improved a new method of DNA extraction from the feather which got high yield as the conventional method. 内容記述 研究概要(和文):キジ亜科間の系統関係については、形態学的研究を初め、卵白リゾチーム、DNAハイブリダイゼーション法及びミトコンドリアの塩基配列を基に解析が行われているが、その結果は異なった系統樹を示してきた。そこで生命維持に不可欠なヘモグロビンの配列を決定し、先の結果と比較検討する事を試みた。最初の計画では、血液の入手が困難であることも考慮しPCRを多用する予定であったが、幸いにも血液が入手できたので、今までのデータの蓄積等を考慮し、まずアミノ酸配列の決定を行い、キジ亜科間の系統関係を明らかにした。その結果、ミトコンドリアの塩基配列とは似ているが、アメリカウズラ亜科とホロホロ鳥亜科の関係が逆転している系統樹を得ることができた。この分子系統樹の結果は、現在得られている化石のデータとも一致しているが、残念な事に、アメリカウズラ亜科の分岐に関する化石は発見されていない。今回得られた系統樹をより確信できるよう、若干のアメリカウズラ亜科の種を加え再検討すべきであると考えている。また、これらの解析等と平行して、稀少種への対応の為に羽よりPCR反応ができないかについて検討を重ねてきた。これまでの抽出方法では、DNAの抽出効率が悪いためミトコンドリアDNAの様な多コピー性の領域は、比較的簡単に増幅することが可能であった。しかしミトコンドリア以外の領域を増幅する為には多少問題があるため。DNA抽出方法を改良し、効率よく抽出できるようになった。そこでこれまでに得られたキジ亜科の羽よりDNAを抽出し、PCR反応により増幅する事が可能になり、塩基配列の決定までを行えるようになった。
