We studied the effects of individual components of a nucleoside-nucleotide mixture on the lymphocyte responses to allonantigens in popliteal lymph node (PLN) of mice. Mice were fed for 4 weeks a nucleotide-free 20% casein diet (control), or this diet supplemented with 3 mol/kg of one of the followings; inosine, guanosine monophosphate (GMP 2Na), uridine, cytidine, thymidine, or a mixture of these. In the graft vs host reaction, there was no difference in the PLN weights and the expression of CD4 and CD8 surface markers among the groups. In the host vs graft reaction there was a significant increase in the PLN weights between the mice fed the supplemented diets and the nucleotide-free dietary group (control). However, there was no difference between the mixture and the individual component diet groups. Except for GMP 2Na and cytidine supplemented groups, there was a difference in the expression of CD4 surface marker in the supplemented groups as compared to the control dietary group; the CD4^+ cells significantly increased in the supplemented groups, although there was no difference between the mixture and individual componenets supplemented groups. We conclude that mice fed the nucleoside-nucleotide mixture and the individual component of the mixture had enhancing effects in the host vs graft responses studied and that the effects in mice fed the mixture were not different from those in mice fed the individual component. |
