Shrinkage of the spleen occurs through contraction of smooth muscles in the capsule and the trabecula responding primarily to stimulation of $ \\alpha $_1-adrenoceptors, leading to mobilization of concentrated blood cells stored in the red pulp into general circulation. Although a variety of endogenous vasoactive substances are found to modify the tone of splenic capsular smooth muscles, the influence of vasoddilators on the splenic contractility has been less investigated as compared with that of vasoconstrictors. Prostacyclin is a powerful vasodilator and inhibitor of platelet aggregation, and relaxes many smooth muscles including bronchial, tracheal, uterus and gastrointestinal muscles besides those of the vasculature. Thus, the present study was conducted to evaluate the effects of prostacyclin, which was given into the splenic artery in order to minimize alterations in systemic hemodynamics, on splenic diameter, vascular tone and mobilization of blood cell from the spleen in anesthetized dogs.
