Thirty-seven strains from several human intestines and four reference strains of B. fragilis were examined for both bacteriocin production and bacteriocin susceptibility. Six bacteriocin-prducing strains were found. Five strains (I-1, 419, 544, 622 and 663) of these strains belong to the B. fragilis ss. fragilis, and one strain(I-1) belongs to the B. fragilis ss. vulgatus. Most of B. fragilis used were sensitive to one or more of the bacteriocins and only one strain(B5, ss. vulgatus) of them was sensitive to all of the bacteriocins isolated. Two strains(I-1 and 622) of those tested were resistant, and they produced bacteriocin. In addition the growth of all of the bacteriocin-producing strains were not inhibited by their own bacteriocin. Host ranges of two bacteriocins produced by strains 0-4 and 622 were broad; thirty-three strains of those tested were inhibited(81%). On the other hand, four bacteriocins(419, 544, 663 and I-1) had a narrow spectrum, respectively. Among B. fragilis subspecies, bacteriocins produced by ss. fragilis were active against all of the subspecies belonging to b. fragilis, but I-1 bacteriocin acted only on ss. vulgatus. This finding is particularly interesting in showing possible application of bacteriocin susceptibility to subspecies classification of B. frabilis. These bacteriocins did not inhibit the growth of the other bacteria expect Bacteroides. Furthermore, crude bacteriocins produced by strains 0-4, 622 and I-1 were compared for some porperties. The largest production of these bacteriocins was obtained under N2 in the brain heart infusion broth-supplemented and GAM broth. The activity of those bacteriocins was detected in culture supernatant, and the highest titer (4-8 A.U.) was obtained in the stationary phase. 0-4 bacteriocin was trypsin-sensitive and heat-resistant, and its inhibition zone was clear. 622 bacteriocin was trypsin-resistant and heat-resistant, but inactivated with heat treatment at 100℃ for 30 minutes. I-1 bacteriocin was less trypsin sensitive and heat-resistant.The latter two bacteriocins gave clear inhibition zone with halo. The other physico-chemical properties of three bacteriocins were similar. |
